Overview

What spermidine trihydrochloride is

Spermidine is a polyamine involved in cell growth, DNA stability and autophagy. As an ingredient it is almost always supplied as the trihydrochloride salt rather than the free base, because the salt is crystalline, non-volatile and far easier to weigh, blend and analyse.

The two CAS numbers trip people up. 124-20-9 is spermidine free base — a viscous, hygroscopic liquid that absorbs CO₂ from air. 334-50-9 is the trihydrochloride we ship. Buyers' specification sheets cite one or the other, and a quotation that matches the wrong number is a specification error waiting to happen, so both appear on ours.

Eastling supplies spermidine trihydrochloride as a white crystalline powder at ≥99% by HPLC, from 1 g for evaluation to kilogram lots. It is synthetic, not a wheat-germ extract — which removes the cadmium question that plant concentrates have to answer.

  • Trihydrochloride salt — crystalline, free-flowing, easy to blend
  • Assay ≥ 99% by HPLC, with water content by Karl Fischer
  • Synthetic, so no plant-derived cadmium load
  • Both CAS numbers documented on the specification

Manufacture

How it is made.

A synthetic route, chosen because it gives a defined assay and avoids the heavy-metal variability of plant concentrates.

  1. 01

    Diamine build-up

Spermidine is assembled from a putrescine-type diamine building block by controlled chain extension — a route that gives the linear polyamine rather than a mixture of homologues.

  2. 02

    Salt formation

The free base is converted to the trihydrochloride in aqueous HCl; the stoichiometry is what fixes the 3:1 counterion ratio your specification refers to.

  3. 03

    Recrystallisation

Recrystallisation from aqueous alcohol sets the crystal form, the colour and the residual solvent profile.

  4. 04

    Drying and release

Vacuum drying to the specified water content, then assay, pH of solution, heavy metals and microbiological testing before release.

Applications

Where it is used.

Spermidine is dosed in milligrams, so the specification matters more than the tonnage.

01

Longevity supplements

Capsules at 1–10 mg, often positioned against wheat-germ extracts. Synthetic material gives a defined dose without the cadmium load that plant concentrates carry.

02

Cell & autophagy research

Media supplementation and assay standards, where the counterion matters because the salt adds three chloride ions per molecule.

03

Functional food

Small inclusion levels in powders and bars, subject to local novel-food rules — check your market before formulating.

Specification

Release criteria.

The written limits we release against, with the method used to test each one. Batch results for the lot you receive are stated on its certificate of analysis.

Tighter limits — for example a lower heavy-metal ceiling or a residual-solvent panel — can be quoted against your own specification wording.

Certificate

Reading the certificate.

Spermidine is usually bought against a competitor's specification sheet. Three lines decide whether the quotation is comparable.

01

Which CAS the assay refers to

A result of 99% as free base is about 57% of the salt's molecular weight in the same powder. Always check whether the assay is expressed on the salt or the base — the difference is not a rounding error.

02

Counterion stoichiometry

Trihydrochloride means three chlorides per molecule. Material that is partly dihydrochloride weighs differently and buffers differently. Titration or ion chromatography settles it; most COAs omit it.

03

Source and cadmium

Wheat-germ extracts can carry cadmium, which is why some markets set a limit for it. Synthetic material starts clean, but ask for the heavy-metal panel either way.

Documentation & packaging

What ships with it.

The paperwork travels with the goods. If your QA team needs an extra declaration, ask before the order is released — some are issued per production lot and cannot be retrofitted.

01

Certificate of analysis

Assay on the salt, water, pH, heavy metals and microbiological results, with the salt form stated explicitly.

02

Analytical data

HPLC chromatogram and MS or NMR identity confirmation.

03

SDS & specification

Safety data sheet, plus the specification the batch was released against — including both CAS numbers.

04

Declarations on request

Non-GMO, allergen, TSE/BSE and origin statements. Vegan and kosher statements are straightforward for synthetic material.

Questions

Before you order.

The questions buyers actually ask about Spermidine. Anything not covered here, ask directly — you will get an answer from someone technical, not a template.

Why do you quote two CAS numbers?
124-20-9 is spermidine free base; 334-50-9 is the trihydrochloride we ship. Buyers' specification sheets cite one or the other, and matching the wrong one is a real specification error. Both appear on our COA so your QA team can reconcile it with your own paperwork.
Why supply the salt instead of the free base?
The free base is a viscous liquid that absorbs CO₂ from the air, which makes weighing and blending unreliable. The trihydrochloride is crystalline, stable and free-flowing. Almost every commercial supplement uses the salt for exactly this reason.
Is it synthetic or extracted from wheat germ?
Synthetic. That gives a defined assay and avoids the cadmium and batch-variability issues of plant concentrates. If your label requires a “natural source” claim, this material will not support it — be clear about that before you formulate.
Is it hygroscopic?
Mildly. Keep it sealed with desiccant at 2–8 °C. Caking in an opened bag is a moisture problem, not a purity problem, but it will ruin your weighing accuracy.
How much should be used per dose?
We do not advise on dosing. Use levels are set by your formulator and, for food, by the permitted level in your market. We supply the specification and the safety data; the dose is your regulatory decision.
Is it stable in solution?
The salt is stable in water at moderate pH, but polyamines are hygroscopic and can absorb CO₂ at high pH. For liquid formats, buffer carefully and generate your own stability data rather than assuming raw-material stability transfers.
What is the minimum order and lead time?
From 1 g for evaluation. Commercial lots are typically 2–3 weeks from written confirmation; kilogram quantities are usually stocked, so ask about current availability.
Can you supply a lower-chloride or different salt form?
Other salts are possible on request but are not catalogue items. Tell us the counterion and the target assay basis, and we will confirm feasibility and lead time before quoting.
